Long non‑coding RNA GAS5 protects against <i>Mycoplasma pneumoniae</i> pneumonia by regulating the microRNA‑222‑3p/TIMP3 axis

Mycoplasma pneumoniae pneumonia (MPP) is a type of induced by M. (MP) infection. The present study investigated the effect long non‑coding RNA growth arrest‑specific 5 (GAS5) in MPP and underlying molecular mechanism this. expression GAS5, microRNA‑222‑3p, (miR‑222‑3p) tissue inhibitor metalloproteinases‑3 (TIMP3) was using reverse transcription‑quantitative PCR. Lipid‑associated membrane protein (LAMP)‑induced THP‑1 cells were used to model MPP. viability LAMP‑induced analyzed an MTT assay. Expression levels interleukin (IL)‑1β, IL‑6 tumor necrosis factor‑α (TNF‑α) pro‑inflammatory cytokines, anti‑inflammatory cytokine heme oxygenase‑1 (HO‑1) measured ELISA. A dual‑luciferase reporter assay assessed associations among miR‑222‑3p TIMP3. GAS5 TIMP3 downregulated upregulated. GAS5‑overexpression increased cells. upregulation decreased IL‑1β, IL‑6, TNF‑α HO‑1 directly interacted with miR‑222‑3p. target or TIMP3‑knockdown reversed promotion on cell as well inhibitory inflammation caused regulating miR‑222‑3p/TIMP3 axis. These results demonstrated potential therapeutic for treatment.
